【摘要】 目的:观察不同浓度碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)引起体外培养的人晶状体上皮细胞迁移和黏着斑激酶(focal adhesion kinase,FAK)mRNA水平的动态变化。方法:体外培养的第3代人晶状体上皮细胞,建立细胞划痕损伤模型,经0,5,10,15μg/L bFGF作用8 ~16h后,电脑图像法测定细胞迁移距离,RT-PCR法检测0,1,4,8, 16h FAK mRNA水平,免疫荧光法检测整合素β1和增殖细胞核抗原表达阳性率的变化。结果:与对照组比,5μg/L组细胞迁移距离和FAK mRNA表达显著增加(P <0.05);10μg/L组细胞整合素β1表达阳性率显著增加(P <0.05);15μg/L细胞迁移距离和FAK mRNA显著降低(P <0.05)。结论:bFGF引起人晶状体上皮细胞迁移距离和FAK mRNA表达的变化。

Effect of bFGF on expression focal adhesion kinase in human lens epithelial cells

Abstract AIM: To observe the effects of different concentration basic fibroblast growth factor (bFGF) on human lens epithelial cells migration and the dynamic changes in focal adhesion kinase (FAK) mRNA level.METHODS: The scratch wound model of the third passage human lens epithelial cells was established and treated with bFGF of different concentrations (0, 5, 10 and 15μg/L) for 8-16 hour. The images of cell migration were quantitatively measured using a computer-assisted videomicroscopic system. The mRNA level of FAK was measured with reverse transcriptase-polymerase chain reaction (RT-PCR) assay at 0, 1, 4, 8 and 16 hour. The positive expression of integrin β1 and proliferating cell nuclear antigen (PCNA) were studied by immunofluorescent staining.RESULTS: The cell migration and FAK mRNA level were significantly stimulated when treated with bFGF at the concentrations of 5μg/L compared with the control group (P <0.05). The integrin β1 positive expression rate was significantly increased when treated with 10μg/L bFGF (P <0.05). The cell migration and FAK mRNA level was significantly inhibited when treated with 15μg/L bFGF (P <0.05). CONCLUSION: The cell migration distance and FAK mRNA expression varies when the cells are treated with different concentration of bFGF. FAK plays an important role in human lens epithelial cells migration.

黏着斑激酶(focal adhesion kinase,FAK)是细胞黏着依赖的信号转导(adhesion-dependent signal transduction )中的酪氨酸蛋白激酶,属于非受体蛋白酪氨酸激酶,在大多组织细胞中皆有表达,参与调节细胞发育、生长、存活、凋亡、黏附、骨架重组等过程,是生长因子受体信号转导通路中的重要信号分子[1,2]。碱性成纤维细胞生长因子(basic fibroblast growth factor, bFGF) 是强效促细胞增殖因子,对细胞的黏附、迁移也有作用 [3,4]。我们探讨不同浓度bFGF引起体外培养的人晶状体上皮细胞损伤后迁移距离与FAK的动态变化,以揭示其对晶状体上皮细胞迁移与增殖的影响。
