[摘要] 目的 观察灯盏花提取物(DSX)对体外高压培养视网膜神经节细胞凋亡及凋亡基因表达的影响。 方法 (1)在RGCs中加入DSX,加压培养后收集细胞,用流式细胞仪测定凋亡细胞百分率。(2)在RGCs中加入DSX,加压培养后收集细胞,用基因芯片寻找凋亡基因的表达。结果 (1)在RGCs高压培养时,DSX组的凋亡百分率低于模型组和对照组。(2)在高压培养的条件下,与神经元细胞凋亡相关的基因S100B表达恢复正常,NGFR表达下调。 结论 DSX是具有RGCs保护作用的有效单体,能使高压培养RGCs的S100B的表达恢复正常,NGFR的表达下调。

Apoptotic genetic expression of RGCs by bolting constituent from Herba Erigerontis cultured in vitro by high pressure

[Abstract] Objective To observe the apoptosis and look for apoptotic gene of RGCs cultured in vitro by high pressure by adding the constituent come from Herba Erigerontis(DSX). Methods We build rat retinal cells and RGCs cultured in virto by high pressure.Put 80 mmHg pressure to RGCs in close tightly culturebottle for four hours, then collect RGCs and check the sum of apoptosis cells by flow cytometry(FCM).Put the pressure 80 mmHg to RGCs in close tightly culturebottle for four hours,then collect RGCs and look for the apoptotic gene by gene chip. Results The apoptosis of model group and control group higher than DSX group cultured in the pressure of 80 mmHg.Two apoptotic gene (S100B,NGFR) express upregulation. Conclusion DSX was the most effective constituent in protecting RGCs in vitro.The expressing upregulation of S100B and NGFR is the direct reasons of the apoptosis of RGCs by pressure.

青光眼系具有病理性高眼压或正常眼压合并视乳头、视网膜神经纤维层损害及青光眼性视野改变的一种致盲性的眼病,目前临床疗效不理想,尤其是控制眼压后,患者视功能仍然继续下降,所以寻找对视神经具有保护作用的药物成为新的要求。DSX是从中药灯盏花中提取的有效成分。目前已证明以DSX为主要组分制备的优视胶囊具有活血化瘀、通窍明目的作用,能保护高眼压状态下的RGCs免受或少受损伤并挽救凋亡细胞[1,2],在此基础上,我们希望能明确其作用机制,为进一步研究的剂型做准备。
