Study of the effects of deep cryopreservation on apoptosis in fetal cornea

Abstract
AIM: To sum the number of apoptotic cells in both normal fetal cornea and the deep cryopreservative fetal cornea and to assess the effects of deep cryopreservation to apoptosis.
METHODS: The TUNEL (TdT mediated biotin dUTP nick-end labeling, TUNEL) technique was used to study if apoptosis was appear in both nomal fetal cornea and cryopreservative cornea in 20 fetal cornea.
RESULTS: The number of apoptotic cells was significantly higher after cryopreservation (7.483±2.490) than that of noncryopreservation (0.817±0.612,P<0.05).
CONCLUSION: Apoptosis is seldom seen in normal fetal cornea but is obvious in deep cryopreservative fetal cornea. That shows freeze-thaw trauma maybe relate to cell apoptosis.

摘要
目的:研究新鲜和深低温保存后的胎儿角膜的细胞凋亡情况。
方法:应用TUNEL法对20例新鲜及深低温保存后胎儿角膜进行了细胞凋亡的研究。
结果:凋亡的角膜细胞全为细胞核着色。新鲜角膜中平均每高倍视野可见凋亡细胞0.817±0.612个,经冷冻保存后的角膜每高倍视野可见凋亡细胞平均为 7.483±2.490个,经统计学处理P<0.05,差异有显著性。
结论:新鲜胎儿角膜可见极少量凋亡细胞。胎儿角膜经深低温冷冻保存后,凋亡细胞数量显著增加。降温-复温损伤可能与细胞凋亡有关。
