The Inhibitory Effect of Diclofenac Sodium on theProliferation of Rabbit Corneal Epithelial Cells in vitro

Purpose:To investigate the inhibitory effect of diclofenac sodium on rabbit corneal epithelial cells(RCECs)in vitro and explore it’s pharmacological mechanism.
Methods: RCECs by culturing the fresh rabbite cornea.Then we selected all second culture of RCECs to research.The second culture of RCECs were exposed to different concentrations of diclofenac sodium in experimental group and to culture media in contrast group.The inhibtive characteristics of diclofenac sodium to the cells’
proliferation were measured with MTT assay.The changes of cells cycles and the apoptosis rate were observed with flow cytometer(FCM).
Results:RCECs cultured with diclofenac sodium showd sinificant inhibition at 24h、48h and 72hof dosage dependence.
Conclusion:Diclofenac sodium has obvious inhibitory effect on RCECs of dosage—dependence.Diclofenac sodium demonstrates antiproliferative effect by inducing cells apoptosis and ceasing cells cycles.
