Purpose:To explore the underlying mechanism of reconstructing damaged corneal surface by epidermal cells.
Method:Engineering tissue combined epidermal cells with human amniotic membrane,then,were transplanted in goats with acute full-thickness skin defect(AFTSD)and limbal stem cell deficiency(LSCD)respectively,and analyzed the p63 expression pattern of regenerated tissue from cornea and skin by confocal microscope.
Results:Results show that△αp63 positive cells clustered in the spinous and granular layers of regenerated epidermis,which were CK 1 negative,suggesting that the dedifferentiation of epidermal cells to stem cells in vivo during the process of tissue regeneration.In addition,groups of△αp63 positive,and CK12 negative cells could be seen in suprabasal layer of reconstructed cornea.
Conclusion:Those findings showed that dedifferentiation of differentiated cells to stem cells in vivo will be attributed to the process of tissue regeneration.
