The expression of CD40 by human retroocular fibroblast

Abstract AIM: To quantify the mRNA expression of CD40 induced by rhIFN-γ in cultured human retroocular fibroblast (RF) from normal subjects and thyroid associated ophthalmopathy (TAO) patients. ETHODS: The mRNA expression of CD40 induced by rhIFN-γ was half quantified in cultured human RF with reverse transcription-polymerase chain reaction (RT-PCR). RESULTS: The absorbency values of 100, 200, 400, 1 000kU/L rhIFN-γ was 0.483±0.095 and 0.476±0.051, 0.647±0.034 and 0.659±0.083, 0.849±0.017and 0.852±0.024, 0.891±0.082 and 0.894±0.041 compared to 0.203±0.032 and 0.213±0.020 of the absorbency values of 0kU/L rhIFN-γ respectively in RF from normal subjects and patients with TAO (P<0.01), with no significant difference at the same concentration (P >0.05). The absorbency values of 12, 24, 48, 72 hours of 400kU/L rhIFN-γ was 0.331±0.047 and 0.329±0.025, 0.668±0.067 and 0.683±0.104, 0.849±0.017 and 0.852±0.024, 0.865±0.021 and 0.876±0.014 compared to 0.203±0.032 and 0.213±0.020 of the absorbency values of 0kU/L rhIFN-γ respectively in RF from normal subjects and patients with TAO (P<0.05), with no significant difference at the same time (P>0.05). CONCLUSION: The cultured human RF from both normal subjects and patients with TAO expresses CD40 mRNA. The rhIFN-γ could up-regulate CD40 gene mRNA expression in human RF from both normal subjects and patients with TAO in a concentration- and time-dependent manner.

摘要目的:探讨rhIFN-γ作用下,正常人和TAO患者球后成纤维细胞(RF)CD40 mRNA的表达情况。方法:用RT-PCR法,对rhIFN-γ诱导正常人和TAO患者RF CD40的mRNA进行半定量分析。结果:经100,200,400,1 000kU/L rhIFN-γ作用48h后,正常人和TAO患者RF CD40的吸光度比值分别为0.483±0.095和0.476±0.051,0.647±0.034和0.659±0.083,0.849±0.017和0.852±0.024,0.891±0.082和0.894±0.041与各自对照组0.203±0.032和0.213±0.020相比,差异有统计学意义(P ≤0.01);两者同一浓度相比差异无统计学意义(P >0.05)。经400kU/L rhIFN-γ作用12,24,48,72h后,正常人和TAO患者RF CD40的吸光度比值分别为0.331±0.047和0.329±0.025,0.668±0.067和0.683±0.104,0.849±0.017和0.852±0.024,0.865±0.021和0.876±0.014与各自对照组0.203±0.032和0.213±0.020相比,差异有统计学意义 (P <0.05),两者同一时间相比差异无统计学意义(P >0.05)。 结论:rhIFN-γ可上调正常人和TAO患者RF CD40 mRNA的水平,呈浓度和时间依赖。
