Altered Proliferation and Expression of TGF-β in Cultured Tenon’s Fibroblast from Glaucoma Patients

Purpose: To evaluate proliferation and the mRNA and protein expression of three TGF-β isoforms in cultured human Tenon’s fibroblast (HTF) in glaucoma patients.
Methods: Proliferation of HTF was investigated with cultured HTF in different concentrations of FBS. The reverse transcriptase-polymerase chain reaction (RT-PCR) was used to detect the mRNA level of three TGF-β isoforms of cultured HTF from glaucoma patients (POAG) and from healthy donors. The intracellular localization of TGF-β2 protein and the expression level of TGF-β1 protein after topical medication treatment in vitro was respectively evaluated by immunohistochemistry and by ELISA.
Results: all three isoforms of TGF-β were expressed on mRNA level by HTF. Compared with control, the expression on mRNA level of TGF-β2 and -β3 were significantly higher in HTF from glaucoma patients,. The TGF-β2 protein expression level was overexpressed in HTF from glaucoma patients. A significantly enhanced cell proliferation (P<0.01) was also found in HTF from glaucoma patients.
Conclusions: The increased proliferation and enhanced expression level of TGF-β, which might support proliferation, might result from prior use of topical medications, which were used by all glaucoma patients in this study for at least one year. And both enhancement of HTF proliferation and TGF-β might have a positive effect on HTF wound healing after antiglaucoma filtration surgery, but might negatively influent surgery result.
