【摘要】 目的:建立一阶导数光谱法测定复方氯霉素滴眼液中氯霉素含量的方法。方法:一阶导数光谱法直接测定复方氯霉素滴眼液中氯霉素含量,于296.3 nm波长处测定振幅D值。结果:氯霉素在5.15 μg/ml~25.75 μg/ml范围呈良好线性关系;其回归方程为D=11.204×10-4C-0.5×10-4(r=0.999 8);平均回收率100.02%、RSD=0.10%(n=5)。结论:本方法操作简单、准确,可用于测定复方氯霉素滴眼液中氯霉素的含量。

Determination of Chloramphenicol in Compound Chloramphenicol Ophthalmic
Solution by First Order Derivative Spectrophotometry

(Beihai People's Hospital,Beihai,Guangxi 536000,China) Abstract:Objective To determine the content of Chloramphenicol in Compound Chloramphenicol ophthalmic Solution.Methods First Order Derivative Spectrophotometry was used.The peak amplitude was determined when the wave length was at 296.3nm.Results The Chloramphenicol showed a good linear relationship within a ranger of 5.15 μg/ml~25.75 μg/ml,the regression equation was D=11.204×10-4C-0.5×10-4 (r=0.999 8);the average recovery was 100.02%and RSD=0.10%(n=5)。Conclusion The method is simple and accurate, and can be used for determination of Chloramphenicol in Compound Chloramphenicol Ophthalmic Solution. Key words:Compound chioramphenicol ophthalmic solution;Chloramphenicol;First order derivative spectrophotometry;Content determination 复方氯霉素滴眼液是由醋酸氢化泼尼松、氯霉素、硼酸、硼砂等组成,具有抗菌消炎、抗过敏等作用,用于过敏性结膜炎、角膜炎、急性虹膜腱状体炎等。本品为复方制剂,由于其他成分的影响不能直接用普通紫外分光光度法测定氯霉素的含量,笔者经过试验后发现,采用一阶导数紫外分光光度法[1]可不经分离直接测定复方氯霉素滴眼液中氯霉素的含量,且结果理想。
1 仪器与试药 仪器UV-1201型双光束紫外可见分光光度计(北京瑞利分析仪器公司),BP21lD型电子天平(德国SARTORIUS);氯霉素对照品(中国药品生物制品检定所提供);复方氯霉素滴眼液(批号:060221)、复方氯霉素滴眼液阴性对照液(批号:060221)(缺氯霉素)及蒸馏水均由本院制剂室提供。无水乙醇(分析纯)。
2 方法与结果
2.1 测定条件及吸收光谱的选择 分别称取氯霉素对照品、复方氯霉素滴眼液及阴性对照液(缺氯霉素)适量,用无水乙醇溶解,稀释成适宜浓度,制成氯霉素对照液(b液),复方氯霉素滴眼液(a液),复方氯霉素滴眼液阴性对照液(缺氯霉素)(c液)在200 nm~400 nm范围内扫描。扫描条件:光度模式为Abs,扫描模式为单一,扫描速度为快速,光谱带宽为2 nm,采样间隔为0.1 nm,见图1。
