【摘要】目的:研究基质细胞衍生因子1α(stromal cellderived factor1α,SDF1α)对体外培养的人类视网膜色素上皮(human retinal pigment epithelium,hRPE)细胞增殖的影响。
方法:应用MTT法研究SDF1α对体外培养的人RPE细胞增殖的影响;用细胞化学染色法检测添加SDF1α前后人视网膜色素上皮细胞内细胞增殖核抗原(proliferating cell nucler antigen,PCNA)的表达情况。
结果:MTT法表明SDF1α可促进体外培养的hRPE细胞的增殖(P<0.05);细胞化学电镜下观察发现:添加SDF1α前hRPE细胞未表达或低表达PCNA,而添加SDF1α后细胞高度表达PCNA。
结论:细胞因子SDF 1α可促进体外培养的hRPE细胞增殖。

Abstract AIM: To investigate the effect of stromal cellderived factor1α (SDF1α) on the proliferation of human retinal pigment epithelium (hRPE) in vitro. METHODS: MTT was used to investigate the effect of SDF1α on the proliferation of hRPE in vitro. Cytochemical staining method was used to detect the expression of proliferating cell nuclear antigen (PCNA) in hRPE with SDF1α. RESULTS: MTT showed that SDF 1α could stimulate the proliferation of hRPE in vitro(P<0.05). Cytochemistry results indicated that there were no or low expression of PCNA before adding SDF1α, but the expression became higher after adding SDF1α observed under electric mirror. CONCLUSION: Cytokine SDF1α can stimulate the proliferation of hRPE cultured in vitro. KEYWORDS: human retinal pigment epithelium; stromal cellderived factor1α; culture in vitro; proliferation
0引言 随着炎症性眼病、白内障的控制和人类寿命的延长,由萎缩、变性和死亡导致的RPE细胞数量减少所致的功能障碍,已经成为很多视网膜疾病如年龄相关性黄斑变性及视网膜色素上皮变性等的原因[1]。寻找如何在体内抑制RPE细胞的丧失或维持RPE细胞功能的方法已成为治疗上述疾病的关键。近年来,视网膜色素上皮移植的兴起可望成为将来治疗这一类疾病的一种新技术[2]。体外传代培养是获取大量人类视网膜色素上皮(human retinal pigment epithelium,hRPE)细胞进行研究和移植的首要条件。分析细胞因子促RPE细胞增殖效应,是体外获取大量的供体RPE来源的重要方法之一。这将为RPE细胞变性性疾病的移植治疗的前景提供新的证据。本文通过研究基质细胞衍生因子1α(stromal cellderived factor1α,SDF-1α)对体外培养的hRPE细胞生长及其DNA合成的促进作用,为进一步明确SDF1α的促RPE细胞增殖作用机制提供实验基础,为临床应用SDF1α治疗各种视网膜变性性疾病提供理论依据。
