摘要 目的 研究复发性单纯疱疹性角膜炎患者血清白细胞介素-2(IL-2)及其可溶性白细胞介素-2受体(SIL-2R)水平。方法 本文测定了30例复发性单纯疱疹性角膜炎患者(实验组)和25例健康人(对照组)血清白细胞介素-2(IL-2)及其可溶性白细胞介素-2受体(SIL-2R)的水平。结果 对照组IL-2的水平为5.41±1.45mg·L-1，实验组IL-2R水平为2.80±1.47mg·L-1，2组比较t=2.90，P＜0.01，说明复发性单纯疱疹性角膜炎患者血清IL-2水平较对照组明显降低。对照组SIL-2R水平为90.86±17.90mg·L-1，实验组SIL-2R水平为171.46±19.30mg·L-1，2组比较t=5.17,P＜0.01。复发性单纯疱疹性角膜炎患者血清SIL-2R水平较对照组显著升高。结论 复发性单纯疱疹性角膜炎患者体内免疫功能紊乱，血清IL-2活性降低、SIL-2R水平升高可能是其原因之一，这也为免疫治疗提供了一定依据。
Study on the levels of serum interleukin-2 and soluble interleukin-2 receptor in patients with recurrent simple herpetic keratitis

Abstract Objective To study the levels of serum interleukin-2 and soluble interleukin-2 receptor(SIL-2R) in recurrent patients with simple herpetic keratitis.Methods The interleukin-2 and soluble interleukin-2 receptor(SIL-2R) in 30 cases of recurrent simple herpetic keratitis experimental group and 25 cases of healthy persons(control group) were determined.Results The serum IL-2 level was 5.41±1.45mg·L-1in normal group,and 2.80±1.47mg·L-1in the patients with recurrent simple herpetic keratitis.The serum IL-2 level in recurrent simple herpetic keratitis was significantly lower than that in normal group(t=2.90,P＜0.01).The serum SIL-2R level was 90.86±17.90mg·L-1 in normal group and 171.46±19.30mg·L-1 in the patients with recurrent simple herpetic keratitis.The serum SIL-2 level in recurrent simple herpetic keratitis was significantly higher than that in normal group(t=5.17,P＜0.01).Conclusions Immune function of recurrent simple herpetic keratitis is lower than that of healthy people.A significant decrease in IL-2 level and a significant increase in SIL-2R level maybe one of causes of this condition.The findings can provide foundation for immunotherapy.

1 材料和方法
1.1 一般资料 观察组：30例(30只眼)单纯疱疹性角膜炎患者(树枝状角膜炎或地图状角膜炎)全部具有复发史，均经裂隙灯显微镜角膜荧光素染色检查确诊，其中男17例，女13例；右眼16例，左眼14例；年龄≤20a者3例，21～40a者12例，41～60a者13例，≥61a者2例；年龄11～67a，平均35.40±15.47a。正常对照组：25例健康人外周血由我院血站提供，25例25只眼，其中男15例，女10例；右眼13例，左眼12例；年龄≤20a者2例，21～40a者11例，41～60a者10例，≥61a者2例；年龄12～65a，平均为34.60±13.57a。
1.2 血清IL-2测定 试剂盒购自长春恩华生物公司，采用夹心ELISA方法，全部操作严格按说明书进行。
1.3 血清SIL-2R测定 试剂盒购自白求恩医科大学免疫室，全部操作严格按说明书进行。采用ELISA双抗夹心法，加抗白细胞介素-2受体(IL-2R)单抗于聚乙烯板(5mg·L-1)4℃ 48h后加封闭液每孔200μL，37℃ 1.5h洗涤加样品稀释液每孔100μL及待测血清50μL混匀。同时设空白对造及标准品系列孔，37℃ 2h洗3遍，各孔加多克隆抗IL-2R多抗100μL、37℃ 2h洗3遍，各孔加驴抗兔抗体100μL、37℃ 2h洗3遍加底物液100μL，10min加50mL终止液，用酶标仪于490nm波长处测OD值，绘制标准曲线，查出SIL-2R含量。
2 结果
2.1 2组血清IL-2及其SIL-2R的水平测定结果，见表1。
