摘要 目的 从蛋白质水平了解体外培养牛眼小梁细胞是否表达转化生长因子β(TGF-β)及其受体(TGF-βR)。方法 采用Supervision法对体外培养第3代牛眼小梁细胞进行TGF-β1,TGF-βR免疫组化检测。结果 小梁细胞TGF-β,-β2和TGF-βRⅠ,RⅡ,RⅢ免疫组化染色的阳性信号分别位于胞浆内和胞膜上,阴性对照未见阳性信号。结论 牛眼小梁细胞表达TGF-β,TGF-βR蛋白。较之猪眼小梁细胞,牛眼小梁细胞更适用于今后研究TGF-β及其受体与原发开角型青光眼发病学的关系。

Expression of TGF-β and TGF-β receptors protein in cultured bovine trabecular meshwork cells

Abstract Objective To study whether cultured bovine trabecular meshwork cells express transforming growth factor-β(TGF-β) and TGF-β receptors(TGF-βR) in protein level.Methods Bovine trabecular meshwork cells were cultured in vitro and passaged 3 times,then the cells were transfered or cultured onto sterile cover lips,and the expressions of TGF-β and TGF-βR were detected with immunohistochemical technique-Supervision method.Results TGF-β1,-β2were significantly expressed in the cytoplasm of cultured bovine trabecular meshwork cells,and TGF-βR Ⅰ,RⅡ,RⅢ immunostain were positive on the membrane of the cells.No immunostaining was seen in the negative control.Conclusion The presence of TGF-β and TGF-βR in cultured bovine trabecular cells were confirmed.Bovine trabecular cells may be more suitable to investigate the relationship between TGF-β,TGF-βR and primary open-angle glaucoma than porcine trabecular cells.

原发开角型青光眼(primary open-angle glaucoma,POAG)是常见致盲性疾病之一。小梁细胞数目异常减少,细胞外基质异常沉积是其主要病理改变[1,2]。有研究表明,具有抑制小梁细胞生长[3,4]、促其细胞外基质合成[5,6]等作用的转化生长因子-β(transforming growth factor-β,TGF-β)在POAG患者房水中的浓度明显高于正常人,可能参与了POAG的发生[7]。本实验拟对体外培养牛眼小梁细胞TGF-β及转化生长因子β受体(transforming growth factor-β receptors,TGF-βR)进行免疫组化研究,以利于进一步研究TGF-β及其受体与POAG发病学的关系。
1 材料和方法
1.1 牛眼小梁细胞体外培养和鉴定参见我科过去报道的方法[8]
1.2 制作细胞爬片
取第3代近融合小梁细胞,消化后传入预置消毒盖玻片的培养皿内,置37℃CO2培养箱中培养至细胞生长近融合。取出盖玻片,用PBS充分浸洗,冷丙酮固定30min后,晾干。
1.3 制作细胞涂片
第3代小梁细胞消化后制成细胞悬液,滴在消毒盖玻片上,自然干燥后,冷丙酮固定,晾干待用。
1.4 免疫组化染色(Supervision法)
将细胞爬片和细胞涂片放入0.5%H2O2中30min,以消除内源性过氧化酶活性。蒸馏水洗3次,滴加山羊血清封闭液,室温下10min,以封闭非特异性抗源。甩去山羊血清,分别滴加兔抗人、大鼠、小鼠TGF-β1,-β2和TGF-βRⅠ,RⅡ,RⅢ单克隆抗体(美国Sigma公司产品),4℃湿盒中过夜。第2天取出后用PBS洗3次,滴加Supervision过氧化物酶-山羊抗兔IgG(武汉Boster公司产品),置37℃湿盒中30min,PBS洗4次,DAB显色,显微镜下观察至出现棕黄色阳性信号,水洗终止反应。苏木素轻度复染。梯度酒精脱水,二甲苯透明,光学树脂封片,镜下观察拍照。阴性对照以PBS代替第1,2抗体,阳性对照分别选用皮肤和乳腺癌标本。
2 结果
牛眼小梁细胞TGF-β及TGF-βR免疫组化染色呈阳性,其中TGF-β1,-β2棕黄色阳性信号位于细胞浆内,而TGF-βRⅠ,RⅡ,RⅢ的阳性信号位于细胞膜上,阴性对照未见阳性信号(图1~6)。
