【摘要】 目的:探讨二肽基肽酶II(dipeptidyl peptidase II,DPPII )在正常大鼠眼球各组织中的免疫表达。方法:应用抗生物素蛋白生物素过氧化物酶复合体法,检测DPPII在正常大鼠眼球各组织中的免疫表达。 结果:发现DPPII在角膜上皮细胞、角膜基质层、角膜内皮细胞、睫状体无色素上皮细胞、虹膜色素上皮细胞、晶状体上皮细胞、晶状体纤维、视网膜神经纤维层、神经节细胞、视网膜色素上皮细胞中免疫染色均呈阳性。结论:DPPII存在于角膜上皮细胞、角膜基质层、角膜内皮细胞、睫状体无色素上皮细胞、虹膜色素上皮细胞、晶状体上皮细胞、晶状体纤维、视网膜神经纤维层、神经节细胞、视网膜色素上皮细胞。

Abstract AIM: To investigate the immune expression of dipeptidyl peptidase II (DPPII) in the normal eyeball tissues. METHODS: The eyes were removed after the rats were sacrificed and postfixed with 40g/L formalin for 20 hours, dehydrated in ethanol, substituted with bezene, and embedded in paraffin wax. Paraffin sections about 4μm thick were cut by using a microtome and mounted on slides. Then the immune expression of DPPII in the normal eyeball tissues were investigated with ABC method.RESULTS: DPPII found in corneal epithelial cells, corneal stroma, corneal endothelial cells, ciliary nonpigment epithelial cells, iris pigment epithelial cells, lens epithelial cells, lens fiber, retinal nerve fiber layer, ganglion cells, retinal pigment epithelium cells showed positive staining.CONCLUSION: DPPII are primarily in the corneal epithelium cells, corneal stroma layer, corneal endothelial cells, ciliary nonpigment epithelial cells, iris pigment epithelial cells, lens epithelial cells, lens fiber, retinal nerve fiber layer, ganglion cells, retinal pigment epithelium cells. 二肽基肽酶II(dipeptidyl peptidase II,DPPII)是属于脯氨酸特异的二肽基肽酶类,是一种活性中心以丝氨酸为主的溶酶体蛋白酶,能水解氮末端为脯氨酸或丙氨酸的寡肽,形成氮末端二肽类产物,是一种外切肽酶[1]。据文献报道,DPPII广泛存在于哺乳动物的各种器官、组织细胞和体液中,如大脑、肾脏、肌肉、生殖器官、造血细胞等[2]。虽然已有学者在角膜、晶状体、视网膜组织中检测到DPPII的活性,但对于DPPII在眼球各组织中,如角膜、虹膜、睫状体、视网膜中存在的具体位置和所发挥的生理功能并不十分清楚。本实验应用抗生物素蛋白生物素过氧化物酶复合体方法检测DPPII在正常Wistar大鼠眼球各组织中的免疫表达,为探讨DPPII在眼组织中的生物学效应提供前期理论基础。
1材料和方法
1.1材料
正常Wistar两性大鼠20只,购于吉林大学医学院校实验动物中心,直接脱颈处死,摘取大鼠眼球,放入40g/L甲醛中固定,用梯度乙醇对眼球组织进行脱水,然后用二甲苯置换组织中的乙醇,后将组织在石蜡浸透,3次,每次30min,60℃,将组织放入新的石蜡中冷却凝固,修块后切片,用切片机切取4μm厚切片。DPPII抗体由吉林大学酶工程实验室从大鼠脾脏提取的DPPII纯品免疫家兔所得的DPPII抗体; ABC免疫染色所用的SABPO(R)试剂盒购于日本东京Nichirei公司,30mL/L H2O2及核染色剂Hematoxillin购于日本京都Nacalai Tesque INC公司,DAB显色液购于武汉博士德公司。 1.2方法
ABC染色主要步骤:①4μm的石蜡切片脱蜡后,依次经过下行梯度酒精脱水;②用30mL/L H2O2室温处理10min,抑制内源性过氧化物酶;蒸馏水洗2min,共3次;③100mL/L山羊正常血清封闭杂蛋白,10min;④用PBS按1∶1000倍稀释DPPII多克隆抗体,第一级抗体反应时间为12h,PBS洗2min,共3次;⑤生物素标记的第二级抗体反应10min;PBS洗2min,共3次;⑥过氧化物酶标记的抗生物素蛋白反应5min,PBS洗2min,共3次;⑦DAB显色5~10min,蒸馏水洗涤;⑧用苏木素进行核染色30s;⑨光学显微镜观察结果。
2结果
DPPII在角膜上皮细胞、角膜基质层和角膜内皮细胞表现为强染色;在睫状体无色素上皮细胞、虹膜色素上皮细胞、视网膜神经纤维层、神经节细胞呈正染色;在晶状体上皮细胞、晶状体纤维及视网膜色素上皮细胞呈弱染色。
