【摘要】 目的 探讨川芎嗪对兔持续性高眼压视神经轴突损伤的保护作用。方法 对36只新西兰白兔眼前房注入α-糜蛋白酶,制作成持续性高眼压模型后,随机等分为高眼压对照组和川芎嗪治疗组,治疗组于高眼压持续7 d始肌注川芎嗪,观察不同时间两组动物球结膜微循环变化及视神经轴突光、电镜结构改变。结果 与高眼压对照组相比,川芎嗪组结膜微循环障碍明显减轻(P<0.01),视神经轴突数目多、直径小,占视神经面积百分比高(P<0.01),且肿胀的视神经轴突结构基本正常。结论 川芎嗪可通过改善微循环状况,对兔高眼压视神经轴突起保护作用。
The protective effect of ligustrazine on optic nerve axons in rabbit eyes with continuous elevated IOP

【Abstract】 Objective To investigate the effects of ligustrazine on optic nerve axons in rabbit eyes with continuous elevated intraocular pressure (IOP). Method Thirty-six NZ rabbits with continuous elevated IOP induced by injection of α-chymotrypsin into anterior chamber were divided into two groups by randomization: treatment and control group. Ten mg/kg ligustrazine injected into the treatment group every day for 3 weeks began on the 7th day after IOP elevated continuously. The changes of conjunctival microcirculation and optic nerve axonal ultrastructure were observed at different time points. Results The destruction of conjunctival microcirculation and axonal ultrastructure was milder in treatment group than that in the control group. In addition, the total number of axons and the percentage of axons in optic nerve area were higher, whereas the axonal diameter was smaller in the treatment group. Conclusion It is suggested that ligustrazine protect optic nerve axons from more damage induced by elevated IOP through improving ocular microcirculation.

青光眼经手术治疗后,多数患者眼压能控制在正常水平,但部分患者视神经损害可继续发展。因此,除降低眼压外,如何保护视神经已成为青光眼防治的重要课题。以往的研究表明,血液流变学改变是原发性开角型青光眼的重要发病因素[1]。因此,从改善微循环角度探讨药物对视神经损害的保护作用,无疑将为青光眼的治疗提供新的途径。目前国内外尚未见报道有关高眼压损伤视神经的保护药物,为此,我们制作了兔持续性高眼压模型,研究了川芎嗪在不同时间对兔眼球结膜微循环的影响,并于光、电镜下观察视神经轴突的改变,以期为青光眼的治疗提供新手段。
材料与方法
一、材料
1.动物:本院动物中心标准健康新西兰大白兔36只,体重2.0~3.0 kg,雌雄不限。
2.药物:无锡市第七制药厂生产的盐酸川芎嗪注射液。
二、方法
1.模型制作:将兔固定于实验架内,0.5%地卡因滴眼2或3次,开睑器开睑,由角膜缘透明区向前房内注入0.5 ml 1∶1 000 α-糜蛋白酶,30 min后测眼压,使眼压持续>50 mm Hg(1 mm Hg=0.133 kPa),如眼压<50 mm Hg可补充注入0.2 ml。
2.动物分组:将36只大白兔随机等分为高眼压对照组和川芎嗪治疗组。治疗组于高眼压持续第7 d开始肌肉注射川芎嗪注射液10 mg/kg,每日2次,
