摘要 目的 研究5-氟尿嘧啶(5-Fu)对兔晶状体上皮细胞(LECs)增殖的影响及眼内毒性。方法 传代兔LECs添加不同质量浓度的5-Fu,作用一定时间后,MTT法测定OD值。选取对LECs增殖有明显抑制效果的质量浓度,进行兔眼前房注射,4天后进行角膜内皮分析和眼内炎性细胞分析。结果 体外实验质量浓度为0.25μg/ml有显著的抑制效果(P<0.05);前房注射的5-Fu达5mg/ml时,表现出角膜内皮形态改变及前节炎性反应,随质量浓度升高,程度加重。结论 5-Fu对兔LECs增殖的体外抑制呈时间依赖性和质量浓度依赖性,前房注射5-Fu产生的眼毒性与其质量浓度正相关。

Study on inhibition of rabbit lens epithelial cells proliferation and intraocular
toxicity by 5-Fu

Abstract Objective To study the anti-proliferation effects of 5-fluorouracil(5-Fu) on cultured rabbit lens epithelial cells (LECs) and its intraocular toxicity.Methods Rabbits’ LECs(3×104/ml) of 2~5 passages were put into 96-well culture plates.0.125-250μg/ml of 5-Fu was added.The culture plates were divided into three groups according to exposure time:A (70min),B(1d),C(continuous exposure until fussion of cells appeared in the control group).After different exposure period,MTT assay was used to test the OD values.Three effective concentrations of 5-Fu in vitro were selected to be injected into anterior chambers of white rabbits.Pathological slides of the eye balls were made 4d later.Analysis of corneal endothelial cells and inflammatory cells counting at the corneal margin were performed.Results Under suitable situation,0.25μg/ml 5-Fu significantly inhibited the proliferation of rabbit LECs in vitro.When injected into the anterior chamber,5mg/ml 5-Fu induced the morphololgical changes of corneal stroma,iris and ciliary body.Conclusions The inhibition on LECs proliferation of rabbits by 5-Fu is time and dose-dependent.Under suitable situation,a concentration of 0.125μg/ml can cause obvious inhibitory effects.The intraocular toxicity of 5-Fu through anterior chamber injection showed positive relation to the concentration.

白内障摘出联合后房型人工晶状体(intraocular lens,IOL)植入术后,残留的晶状体上皮细胞(lens epithelial cells,LECs)向后囊表面增殖和移行在后囊混浊的发生过程中起着重要作用。5-氟尿嘧啶(5-fluorouracil,5-Fu)是胸苷酸合成酶的抑制剂,可有效抑制细胞的有丝分裂,寻找5-Fu的适当质量浓度和给药途径,使其有效抑制LECs增殖又不产生眼毒性,对于临床上预防后囊混浊将有重要的指导意义。
1 材料与方法
1.1 体外实验
兔LECs原代细胞取自健康白色家兔。用含有15%胎牛血清(中国医学科学院血液病研究所)的DMEM培养液(美国SIGMA公司)培养传代3~5代后按3×104/ml密度接种于96孔板,贴壁24h。分别添加0.125~250μg/ml的5-Fu,每质量浓度设8孔,对照组为空白培养液,设8孔,各96孔板按作用时间不同分A(70min)、B(24h)、C(作用至对照组细胞融合)3组,其中A,B组终止药物作用后,换液继续培养至对照组细胞融合;采用MTT(美国SIGMA公司)法,测定各孔吸光度(OD)值,计算细胞密度的显著性差异。
1.2 体内实验
健康白色家兔,按50mg/kg静脉慢推氯胺酮麻醉后,分别向前房内注射0.5μg/ml,5mg/ml,25mg/ml的5-Fu0.2~0.3ml,4天后处死家兔,行眼球摘除术。环形剪下角膜,避开房角结构,将角膜内皮细胞行苔盼兰、茜素红复合染色,低倍下照相,照片用细胞分析仪分析内皮形态,去除角膜的眼球制作常规石蜡病理切片,镜下进行角膜缘、虹膜、睫状体炎性细胞分类计数。
1.3 统计学分析
体外实验各96孔板的比色结果分别进行显著性检验:每板每种质量浓度的8个OD值分别和对照组的8个OD值进行t检验;体内实验将各眼球病理切片分别进行10个高倍视野(×400)内炎细胞计数,结果进行t检验。P<0.05差异有显著性。
