【摘要】 目的:研究0.04%倍诺喜(Benoxil)兔前房内注射对角膜内皮细胞的影响。方法:取18只成年健康家兔,随机分为2组,0.04%倍诺喜组(实验组)和平衡盐溶液组(对照组),每组9只(18只眼),前房内注射前及注射后1 d、3 d、14 d,两组双眼均行接触性角膜内皮显微镜测量角膜内皮细胞数、细胞密度、细胞平均面积、面积变异系数(CV值)、六角形细胞百分率并取14 d角膜标本制作电镜切片行形态学观察。结果:0.04%倍诺喜前房注射后3 d,上述5项定量指标与对照组相比差别有统计学意义(P<0.05),14 d时与对照组相比差别无统计学意义(P>0.05),且内皮细胞形态结构与正常及对照组相比变化不明显。结论:0.04%倍诺喜兔前房内注射后3 d对角膜内皮细胞造成了损害,至14 d时,这种影响消失。

Experimental study of Benoxil on the corneal endothelium

Abstract Objective:To study the effects of the corneal endothelium of rabbit after being injected 0.04% Benoxil into the anterior chamber.Methods:Eighteen adult rabbits were randomly divided into Benoxil group and control group respectively,nine rabbits(18 eyes)for each group.All rabbits were examined under the contact endothelial specular microscope for endothelial cell number,cell density,average cell area,coefficient of variation of cell area(CV) and ratio of hexagonal cells before being injected and at 1 day,3 days and 14 days after being injected.The corneal endothelium were observed by transmission electron microscopy at 14 days after being injected.Results:There was not almost changes of the ultrastructural for Benoxil after being injected.In 3 days,there was a significant change of the five quantitative indexes between Benoxil and control group(P<0.05).There was no significant difference in 14 days after being injected in the group and control group(P>0.05).Conclusion:0.04% Benoxil can cause damage of corneal endothelial cells in 3 days after being injected.But the changes will almost recover in 14 days after being injected.

近年来,随着眼科手术技巧及设备的改进,表面麻醉已广泛应用于各种内眼手术并且取得了较好的临床效果[1]。做为常用的表面麻醉剂,倍诺喜的应用与日俱增。但内眼手术存在着由手术切口渗入前房内少量表面麻醉剂的可能,这些麻醉剂的存在是否会对角膜内皮细胞产生不良影响。本实验通过对0.04%倍诺喜兔前房内注射,探讨不同时间角膜内皮细胞的变化情况,报告如下:
1 资料与方法
1.1 一般资料
健康成年家兔18只,体重2.0~2.5Kg,雌雄兼用,无眼疾,由山西医科大学实验动物中心提供。
1.2 实验方法
18只家兔分时间段行双眼测量角膜内皮细胞数、细胞平均面积、细胞密度、面积变异系数(CV)、六角形细胞百分率,250 g/L(4 mL/kg)乌拉坦耳缘静脉麻醉后,4号半的一次性皮试针头在颞侧角膜缘内穿刺入前房,轻微摆动针头排出0.2 mL房水,然后注入0.2%利多卡因0.2 mL,于注射后1 d、3 d、14 d再次测量上述指标,进行统计分析,并于14 d时取角膜标本制作电镜切片行形态学观察。
1.3 统计学分析
结果用±s表示,统计分析采用t检验,P<0.05作为显著标准。
